Krabbe disease is a fatal brain disorder in which the body lacks a critical enzyme needed to maintain the protective coating around nerve cells. The only available treatment — umbilical cord blood transplant — can slow brain deterioration but leaves the nerves outside the brain largely unprotected, allowing the disease to continue damaging the body’s broader nervous system.
This University of Pittsburgh technology pairs cord blood transplantation with gene therapy (delivered via an AAV viral vector) to restore the missing enzyme throughout the body. The two therapies work together: the cord blood rebuilds the immune system and supplies the enzyme to the brain, while the gene therapy ensures the rest of the nervous system is also covered — something no current treatment achieves.
In preclinical studies, the combination significantly extended survival and restored nerve insulation in both the brain and body compared to either therapy alone. Initial human clinical trial data has demonstrated safety and effectiveness.
